{"id":1880,"date":"2026-05-29T09:56:50","date_gmt":"2026-05-29T07:56:50","guid":{"rendered":"https:\/\/www.uni.lu\/lcsb-fr\/?post_type=news&#038;p=1880"},"modified":"2026-05-29T09:56:51","modified_gmt":"2026-05-29T07:56:51","slug":"observer-les-neurones-au-microscope-a-super-resolution","status":"publish","type":"news","link":"https:\/\/www.uni.lu\/lcsb-fr\/news\/observer-les-neurones-au-microscope-a-super-resolution\/","title":{"rendered":"Observer les neurones au microscope \u00e0 super-r\u00e9solution"},"content":{"rendered":"<div class=\"wp-block-unilux-blocks-spacer is-spacer-size-md\"><\/div>\n\n\n<section class=\"py-0 wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>Au LCSB, l\u2019\u00e9quipe <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/chemical-molecular-neurobiology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Chemical and Molecular Neurobiology<\/a> dirig\u00e9e par la <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/ivana-nikic-spiegel\/\" target=\"_blank\" rel=\"noreferrer noopener\">professeure Ivana Niki\u0107-Spiegel<\/a> allie microscopie et biochimie afin de d\u00e9chiffrer les m\u00e9canismes mol\u00e9culaires \u00e0 l&rsquo;origine des maladies du cerveau, plus particuli\u00e8rement la scl\u00e9rose en plaques. Leur objectif : trouver des moyens de prot\u00e9ger les neurones. Leur approche : utiliser l&rsquo;imagerie nanoscopique et des outils mol\u00e9culaires de pointe pour mieux comprendre la neurod\u00e9g\u00e9n\u00e9rescence.<\/p>\n\n\n\n<h2 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"coup-de-projecteur-sur-la-sclerose-en-plaques\"\n    >\nCoup de projecteur sur la scl\u00e9rose en plaques<\/h2>\n\n\n\n<p>L\u2019\u00e9quipe se concentre sur la scl\u00e9rose en plaques, une maladie neurologique chronique qui touche 2,8 millions de personnes dans le monde \u2013 principalement des jeunes adultes \u2013 et se manifeste par des sympt\u00f4mes tr\u00e8s h\u00e9t\u00e9rog\u00e8nes. Certains traitements permettent de retarder la progression de la maladie sur le long terme mais il n\u2019existe actuellement aucun traitement curatif, ce qui a un lourd impact \u00e0 la fois sur les patients et sur les syst\u00e8mes de sant\u00e9. \u00ab La scl\u00e9rose en plaques se caract\u00e9rise par la formation de l\u00e9sions dans le cerveau et la moelle \u00e9pini\u00e8re. Celles-ci r\u00e9sultent d\u2019une inflammation, provoqu\u00e9e par les cellules immunitaires, qui endommage la my\u00e9line, la gaine protectrice des fibres nerveuses, et peut \u00e9galement l\u00e9ser les fibres nerveuses elles-m\u00eames \u00bb, explique Prof. Niki\u0107-Spiegel. \u00ab En cons\u00e9quence, la communication au sein du syst\u00e8me nerveux est perturb\u00e9e, ce qui entra\u00eene l\u2019apparition de diff\u00e9rents sympt\u00f4mes au fil du temps. Les m\u00e9canismes mol\u00e9culaires impliqu\u00e9s dans ce processus ne sont pas encore bien compris. \u00bb<\/p>\n\n\n\n<p>C\u2019est l\u00e0 qu\u2019intervient l\u2019expertise technique unique de cette \u00e9quipe. En utilisant \u00e0 la fois l\u2019imagerie, de la microscopie sur cellules vivantes \u00e0 la microscopie \u00e0 super-r\u00e9solution, la biochimie et la prot\u00e9omique, les chercheurs \u00e9tudient l\u2019organisation des prot\u00e9ines au sein des cellules du cerveau. En \u00ab&nbsp;\u00e9tiquetant&nbsp;\u00bb les prot\u00e9ines nouvellement synth\u00e9tis\u00e9es, dans un type de cellule donn\u00e9 et pendant une p\u00e9riode sp\u00e9cifique, ils sont en mesure d\u2019identifier les m\u00e9canismes perturb\u00e9s par la maladie. \u00ab Nous menons d\u2019abord ce qu\u2019on appelle des \u00e9tudes prot\u00e9omiques non cibl\u00e9es afin de d\u00e9terminer quelles prot\u00e9ines sont affect\u00e9es au cours des diff\u00e9rentes phases de la scl\u00e9rose en plaques. Les filaments du cytosquelette sont visibles gr\u00e2ce \u00e0 un marquage fluorescent.        <\/p>\n    <\/figure><\/div><\/div>\n\n\n\n<div class=\"wp-block-unilux-blocks-wrapper w-full lg:w-5\/12 lg:order-1 lg:px-32\">\n<p>Une fois les prot\u00e9ines prometteuses identifi\u00e9es, les chercheurs ont recours \u00e0 la microscopie de pointe pour \u00e9tudier de plus pr\u00e8s ces nouvelles cibles potentielles ainsi que certaines prot\u00e9ines d\u00e9j\u00e0 connues pour leur r\u00f4le dans la maladie. Ils \u00e9tudient par exemple les canaux ioniques pr\u00e9sents dans les interstices entre les gaines de my\u00e9line et les prot\u00e9ines du cytosquelette qui conf\u00e8rent leur structure aux neurones.<br> <br>L\u2019imagerie de cellules vivantes, combin\u00e9e \u00e0 un marquage fluorescent, leur permet par exemple de suivre la mani\u00e8re dont ces prot\u00e9ines se r\u00e9organisent au fil du temps. Gr\u00e2ce \u00e0 la microscopie \u00e0 super-r\u00e9solution, ils peuvent par ailleurs travailler \u00e0 l\u2019\u00e9chelle d\u2019une seule mol\u00e9cule, ce qui leur permet de cartographier tr\u00e8s pr\u00e9cis\u00e9ment la localisation des prot\u00e9ines au sein des cellules.<\/p>\n\n\n\n<ul class=\"wp-block-unilux-blocks-custom-buttons btn-list\"><\/ul>\n<\/div>\n<\/div>\n<\/div>\n<\/section>\n\n\n<section class=\"py-0 wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>Comme la communication entre les neurones d\u00e9pend de la distribution des prot\u00e9ines \u00e0 l\u2019\u00e9chelle nanom\u00e9trique, cette technologie est essentielle pour d\u00e9tecter des changements subtils qui ne peuvent \u00eatre observ\u00e9s avec l\u2019imagerie conventionnelle. Un tout nouveau microscope r\u00e9cemment install\u00e9 au LCSB permet \u00e0 l\u2019\u00e9quipe de visualiser des prot\u00e9ines ou des agr\u00e9gats de prot\u00e9ines mesurant seulement 10 \u00e0 15 nanom\u00e8tres, ouvrant de nouvelles perspectives pour mieux comprendre comment les alt\u00e9rations de l\u2019organisation mol\u00e9culaire contribuent au dysfonctionnement neuronal.<\/p>\n\n\n\n<h2 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"trouver-des-points-communs-entre-les-maladies-neurodegeneratives\"\n    >\nTrouver des points communs entre les maladies neurod\u00e9g\u00e9n\u00e9ratives<\/h2>\n\n\n\n<p>En exploitant la puissance de ces technologies de pointe et en d\u00e9veloppant de nouvelles m\u00e9thodologies pour leur utilisation dans des syst\u00e8mes complexes tels que les cultures cellulaires et les mod\u00e8les <em>in vivo<\/em>, l\u2019\u00e9quipe vise non seulement \u00e0 \u00e9lucider les m\u00e9canismes mol\u00e9culaires de la scl\u00e9rose en plaques mais aussi \u00e0 trouver des points communs avec d\u2019autres maladies du cerveau.<br><br>\u00ab Des processus tels que le stress oxydatif et la neuro-inflammation jouent un r\u00f4le dans tous les maladies neurod\u00e9g\u00e9n\u00e9ratives.
