Il est donc essentiel de mieux comprendre comment les agr\u00e9gats de prot\u00e9ines toxiques se d\u00e9placent entre les r\u00e9gions du cerveau pour \u00e9lucider les m\u00e9canismes impliqu\u00e9s dans la progression de la maladie, notamment aux stades pr\u00e9coces. Des chercheurs du <a href=\"https:\/\/www.uni.lu\/lcsb-fr\/\" target=\"_blank\" rel=\"noreferrer noopener\">Luxembourg Centre for Systems Biomedicine<\/a> (LCSB) et des collaborateurs internationaux viennent justement d\u2019observer la propagation progressive d\u2019agr\u00e9gats d&rsquo;alpha-synucl\u00e9ine dans des assemblo\u00efdes c\u00e9r\u00e9braux obtenus \u00e0 partir de cellules de patients. Cette \u00e9tude, <a href=\"https:\/\/advanced.onlinelibrary.wiley.com\/doi\/full\/10.1002\/advs.202409040\" target=\"_blank\" rel=\"noreferrer noopener\">publi\u00e9e dans <em>Advanced Science<\/em><\/a>, apporte de nouvelles informations sur la mani\u00e8re dont la maladie de Parkinson progresse d\u2019une zone du cerveau \u00e0 une autre.<\/p>\n\n\n\n<h4 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"etudier-la-propagation-des-agregats-de-proteines\"\n    >\n<strong><strong>\u00c9tudier la propagation des agr\u00e9gats de prot\u00e9ines<\/strong><\/strong><\/h4>\n\n\n\n<p>Si la maladie de Parkinson est associ\u00e9e \u00e0 des sympt\u00f4mes tels que les tremblements, la rigidit\u00e9 musculaire et le ralentissement des mouvements, elle d\u00e9bute en fait de fa\u00e7on silencieuse, bien avant l&rsquo;apparition de ces sympt\u00f4mes moteurs. La pr\u00e9sence de sympt\u00f4mes non moteurs pr\u00e9coces a conduit les chercheurs \u00e0 s&rsquo;interroger sur la mani\u00e8re dont la maladie se propage dans le syst\u00e8me nerveux au fil du temps. L&rsquo;une des th\u00e9ories principales, propos\u00e9e pour la premi\u00e8re fois par l&rsquo;anatomiste allemand Heiko Braak, sugg\u00e8re que la maladie de Parkinson pourrait commencer au niveau l&rsquo;intestin ou du le tronc c\u00e9r\u00e9bral inf\u00e9rieur, avant de remonter progressivement vers le cerveau selon un sch\u00e9ma sp\u00e9cifique. Toutefois, les m\u00e9canismes pr\u00e9cis derri\u00e8re cette propagation restent inconnus.<\/p>\n\n\n\n<p>Une prot\u00e9ine appel\u00e9e alpha-synucl\u00e9ine est au c\u0153ur de la maladie. Dans un cerveau en bonne sant\u00e9, elle contribue \u00e0 r\u00e9guler la communication entre les neurones. Dans la maladie de Parkinson, cette prot\u00e9ine se replie mal et s&rsquo;agr\u00e8ge, endommageant les cellules nerveuses et formant des structures anormales, connues sous le nom de corps de Lewy, caract\u00e9ristiques de la maladie. Il est essentiel de comprendre comment ces agr\u00e9gats se propagent entre les diff\u00e9rentes r\u00e9gions du cerveau pour mettre au point de nouvelles strat\u00e9gies th\u00e9rapeutiques. Pour relever ce d\u00e9fi, l\u2019\u00e9quipe <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/developmental-cellular-biology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Developmental &amp; Cellular Biology<\/a> du LCSB, dirig\u00e9e par le professeur Jens Schwamborn, a collabor\u00e9 avec des chercheurs de l&rsquo;Universit\u00e9 technique d&rsquo;Eindhoven, de l&rsquo;h\u00f4pital universitaire Schleswig-Holstein \u00e0 L\u00fcbeck et de l&rsquo;Institut Pasteur hell\u00e9nique d&rsquo;Ath\u00e8nes pour recr\u00e9er les premiers stades de la maladie de Parkinson en laboratoire \u00e0 l&rsquo;aide d&rsquo;un outil innovant : les assemblo\u00efdes c\u00e9r\u00e9braux.<\/p>\n\n\n\n<h4 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"un-nouveau-modele-pour-la-recherche-sur-la-maladie-de-parkinson\"\n    >\n<strong><strong>Un nouveau mod\u00e8le pour la recherche sur la maladie de Parkinson<\/strong><\/strong><\/h4>\n\n\n\n<p>Pour \u00e9tudier la progression de la maladie, les chercheurs ont mis au point des assemblo\u00efdes en fusionnant deux types d&rsquo;organo\u00efdes c\u00e9r\u00e9braux&nbsp;: des structures tridimensionnelles miniatures obtenues en laboratoire \u00e0 partir de cellules souches humaines. Un des deux organo\u00efdes est un mod\u00e8le du cerveau post\u00e9rieur, la r\u00e9gion o\u00f9 on pense que la maladie de Parkinson d\u00e9bute, et le deuxi\u00e8me repr\u00e9sente le m\u00e9senc\u00e9phale, la zone o\u00f9 se trouvent les neurones producteurs de dopamine qui sont d\u00e9truits par la maladie.<\/p>\n\n\n\n<p>En utilisant des cellules souches pluripotentes induites issues de patients pr\u00e9sentant une triplication rare du g\u00e8ne SNCA, une mutation qui entra\u00eene une surproduction d&rsquo;alpha-synucl\u00e9ine, les chercheurs ont constat\u00e9 que les organo\u00efdes repr\u00e9sentant&nbsp;le cerveau post\u00e9rieur d\u00e9veloppaient spontan\u00e9ment des agr\u00e9gats de prot\u00e9ines toxiques. Lorsque ces organo\u00efdes ont \u00e9t\u00e9 reli\u00e9s aux organo\u00efdes sains repr\u00e9sentant le m\u00e9senc\u00e9phale pour former des assemblo\u00efdes, les agr\u00e9gats d&rsquo;alpha-synucl\u00e9ine ont commenc\u00e9 \u00e0 se r\u00e9pandre dans le tissu c\u00e9r\u00e9bral sain en l&rsquo;espace de 30 jours seulement.<\/p>\n\n\n\n<p>\u00ab Ce mod\u00e8le d&rsquo;assemblo\u00efde obtenu \u00e0 partir de cellules de patients nous donne un aper\u00e7u sans pr\u00e9c\u00e9dent de la mani\u00e8re dont la forme toxique de l&rsquo;alpha-synucl\u00e9ine s&rsquo;accumule et se propage entre les r\u00e9gions du cerveau \u00bb, souligne le <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/jens-christian-schwamborn\/\" target=\"_blank\" rel=\"noreferrer noopener\">professeur Jens Schwamborn<\/a>. \u00ab C&rsquo;est la premi\u00e8re fois que nous avons pu observer la progression de ces agr\u00e9gats dans un mod\u00e8le humain sans avoir \u00e0 surexprimer certaines prot\u00e9ines ou \u00e0 provoquer leur agr\u00e9gation de fa\u00e7on artificielle.&nbsp;\u00bb L&rsquo;\u00e9tude vient donc soutenir l&rsquo;hypoth\u00e8se de Braak en d\u00e9montrant que les agr\u00e9gats toxiques d&rsquo;alpha-synucl\u00e9ine peuvent se propager d&rsquo;une r\u00e9gion c\u00e9r\u00e9brale \u00e0 l&rsquo;autre.<\/p>\n\n\n\n<h4 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"lalpha-synucleine-et-son-role-dans-la-progression-de-la-maladie\"\n    >\n<strong><strong>L&rsquo;alpha-synucl\u00e9ine et son r\u00f4le dans la progression de la maladie<\/strong><\/strong><\/h4>\n\n\n\n<p>L&rsquo;\u00e9tude a ainsi montr\u00e9 qu&rsquo;un exc\u00e8s d&rsquo;alpha-synucl\u00e9ine dans le cerveau post\u00e9rieur \u00e9tait suffisant pour d\u00e9clencher sa propagation vers le m\u00e9senc\u00e9phale et avoir un impact sur les synapses, les points de communication vitaux entre les neurones. En effet, avant m\u00eame que les neurones ne commencent \u00e0 mourir, les chercheurs ont observ\u00e9 des signes pr\u00e9coces de dysfonctionnement synaptique, une \u00e9tape cl\u00e9 de la neurod\u00e9g\u00e9n\u00e9rescence.<\/p>\n\n\n\n<p>\u00ab L\u2019augmentation de la quantit\u00e9 d&rsquo;alpha-synucl\u00e9ine dans le cerveau post\u00e9rieur a d\u00e9clench\u00e9 la propagation de la forme toxique de la prot\u00e9ine vers le m\u00e9senc\u00e9phale sain, entra\u00eenant un dysfonctionnement synaptique \u00bb, d\u00e9taille le professeur Schwamborn. \u00ab Cela montre que les assemblo\u00efdes c\u00e9r\u00e9braux constituent un bon mod\u00e8le pour \u00e9tudier le processus de propagation et pour d\u00e9velopper des approches th\u00e9rapeutiques inhibant ce processus \u00bb.<\/p>\n\n\n\n<p>Dans ces assemblo\u00efdes, les scientifiques ont par ailleurs utilis\u00e9 des techniques de pointe telles que les r\u00e9seaux de micro\u00e9lectrodes et le tra\u00e7age viral pour s\u2019assurer que les deux organo\u00efdes formaient des connexions fonctionnelles et directionnelles similaires \u00e0 ce qui s\u2019observe entre les diff\u00e9rentes r\u00e9gions du cerveau. \u00c0 mesure que l&rsquo;alpha-synucl\u00e9ine se propageait, ces connexions s&rsquo;affaiblissaient, indiquant bien une perturbation pr\u00e9coce de l&rsquo;activit\u00e9 c\u00e9r\u00e9brale.<\/p>\n\n\n\n<h4 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"vers-une-detection-precoce-et-une-medecine-personnalisee\"\n    >\n<strong><strong>Vers une d\u00e9tection pr\u00e9coce et une m\u00e9decine personnalis\u00e9e<\/strong><\/strong><\/h4>\n\n\n\n<p>Les assemblo\u00efdes constituent donc un environnement id\u00e9al pour \u00e9tudier la progression de la maladie de Parkinson avec un niveau de d\u00e9tail sans pr\u00e9c\u00e9dent. En imitant l&rsquo;architecture et la connectivit\u00e9 du cerveau, ils devraient permettent de lever le voile sur les premiers stades de la maladie de Parkinson et de d\u00e9velopper de nouvelles approches pour la m\u00e9decine de pr\u00e9cision. Comme chaque assemblo\u00efde est obtenu \u00e0 partir de cellules d\u2019un patient donn\u00e9, les chercheurs peuvent en effet \u00e9tudier l\u2019impact du patrimoine g\u00e9n\u00e9tique du patient sur le d\u00e9veloppement de la maladie et tester des traitements cibl\u00e9s.<\/p>\n\n\n\n<p>\u00ab Ce mod\u00e8le nous donne l\u2019opportunit\u00e9 d&rsquo;\u00e9tudier les m\u00e9canismes pr\u00e9coces de la maladie et de chercher des traitements susceptibles de bloquer la propagation de l&rsquo;alpha-synucl\u00e9ine avant que des dommages irr\u00e9versibles ne se produisent \u00bb, explique le professeur Schwamborn. \u00ab \u00c0 terme, cela pourrait d\u00e9boucher sur des traitements pr\u00e9ventifs pour les personnes \u00e0 risque qui stopperaient la maladie avant
