Le LCSB se joint ainsi \u00e0 l\u2019<a href=\"https:\/\/alan.lu\/fr\/\" target=\"_blank\" rel=\"noreferrer noopener\">Association luxembourgeoise pour les maladies rares<\/a>&nbsp;(ALAN) et \u00e0 de nombreux b\u00e2timents mis en lumi\u00e8re au Luxembourg et de par le monde pour leur rendre hommage.<\/p>\n\n\n\n<p>Parmi les plus de 6000 maladies rares connues, beaucoup se d\u00e9clarent d\u00e8s l&rsquo;enfance, sont graves voire incurables, et ont un profond impact sur les patients et leurs familles. Cette Journ\u00e9e mondiale des maladies rares nous rappelle qu\u2019il est faut continuer d\u2019investir dans la recherche afin de d\u00e9velopper des traitements efficaces. Au LCSB, le groupe&nbsp;<a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/enzymology-metabolism\/\" target=\"_blank\" rel=\"noreferrer noopener\">Enzymology and Metabolism<\/a>, dirig\u00e9 par le&nbsp;<a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/carole-linster\/\" target=\"_blank\" rel=\"noreferrer noopener\">professeur Carole Linster,<\/a>&nbsp;se place au premier plan en \u00e9tudiant les m\u00e9canismes g\u00e9n\u00e9tiques et mol\u00e9culaires \u00e0 l&rsquo;origine de maladies m\u00e9taboliques rares.&nbsp;<\/p>\n\n\n\n<h4 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"faire-progresser-la-recherche-sur-les-maladies-neuro-metaboliques-rares\"\n    >\n<strong>Faire progresser la recherche sur les maladies neuro-m\u00e9taboliques rares<\/strong><\/h4>\n\n\n\n<p>L\u2019 \u00e9quipe utilise des techniques de pointe pour mieux comprendre certaines maladies neuro-m\u00e9taboliques rares et explorer de possibles strat\u00e9gies th\u00e9rapeutiques. En collaboration avec des partenaires internationaux, ils ont par exemple&nbsp;<a href=\"https:\/\/www.uni.lu\/fr\/news\/des-mutations-genetiques-a-lorigine-dune-nouvelle-maladie-infantile\/\" target=\"_blank\" rel=\"noreferrer noopener\">identifi\u00e9 une maladie infantile rare<\/a>, baptis\u00e9e PEBEL, qui r\u00e9sulte de mutations sur les g\u00e8nes NAXD ou NAXE. Ces mutations perturbent les m\u00e9canismes qui, \u00e0 l\u2019instar d\u2019une \u00e9quipe de maintenance, permettent normalement aux cellules de r\u00e9parer ou d\u2019\u00e9liminer les compos\u00e9s toxiques. Dans ce cas, le processus \u00e9choue et les substances nocives s&rsquo;accumulent, entra\u00eenant les l\u00e9sions c\u00e9r\u00e9brales progressives et&nbsp;les autres sympt\u00f4mes observ\u00e9s chez les jeunes patients.&nbsp;<\/p>\n\n\n\n<p>Des&nbsp;<a href=\"https:\/\/academic.oup.com\/brain\/article\/145\/5\/e41\/6540470\" target=\"_blank\" rel=\"noreferrer noopener\">travaux de recherche<\/a>&nbsp;men\u00e9s par l&rsquo;\u00e9quipe ont permis de d\u00e9couvrir que la niacine, une forme de vitamine B3, pouvait avoir un effet b\u00e9n\u00e9fique sur certains des sympt\u00f4mes li\u00e9s \u00e0 ces mutations g\u00e9n\u00e9tiques. Ce premier r\u00e9sultat soulignant le potentiel des interventions nutritionnelles et de ce qu\u2019on appelle des m\u00e9dicaments \u00e0 petites mol\u00e9cules pour la prise en charge des maladies rares, Prof. Carole Linster et son \u00e9quipe ont pouss\u00e9 plus loin.<\/p>\n\n\n\n<p>Des r\u00e9sultats r\u00e9cents, publi\u00e9s dans la revue scientifique&nbsp;<a href=\"https:\/\/cmbl.biomedcentral.com\/articles\/10.1186\/s11658-024-00681-8\" target=\"_blank\" rel=\"noreferrer noopener\"><em>Cellular &amp; Molecular Biology Letters<\/em><\/a>, ont ainsi confirm\u00e9 que des produits toxiques issus du m\u00e9tabolisme, l\u2019ensemble des r\u00e9actions chimiques qui ont lieu dans l\u2019organisme, s&rsquo;accumulent dans les cellules d\u00e9pourvues de l&rsquo;enzyme NAXD. Ils ont \u00e9galement r\u00e9v\u00e9l\u00e9 que ces m\u00e9tabolites toxiques bloquent l&rsquo;activit\u00e9 d&rsquo;une autre enzyme importante, la 3-phosphoglyc\u00e9rate d\u00e9shydrog\u00e9nase, qui joue un r\u00f4le cl\u00e9 dans la synth\u00e8se de la s\u00e9rine. En l&rsquo;absence d&rsquo;une quantit\u00e9 suffisante de s\u00e9rine, de nombreux processus cellulaires sont perturb\u00e9s, ce qui pourrait contribuer \u00e0 expliquer les sympt\u00f4mes observ\u00e9s chez les enfants atteints de PEBEL. Les chercheurs ont de plus d\u00e9couvert que ce ph\u00e9nom\u00e8ne s&rsquo;aggrave lorsque les cellules sont expos\u00e9es \u00e0 un stress m\u00e9tabolique.&nbsp;<\/p>\n\n\n\n<p>Plus important, l&rsquo;\u00e9quipe a trouv\u00e9 un moyen d&rsquo;aider les cellules malades : des apports compl\u00e9mentaires en nicotinamide riboside, une autre forme de vitamine B3, et en inosine ont partiellement restaur\u00e9 la production de s\u00e9rine et am\u00e9lior\u00e9 les fonctions cellulaires. \u00ab Si nous avons confirm\u00e9 l&rsquo;effet b\u00e9n\u00e9fique connu de la vitamine B3, nous avons constat\u00e9 que la nicotinamide et d&rsquo;autres suppl\u00e9ments, comme l&rsquo;inosine, pouvaient avoir un effet encore plus important, soit en remplacement de la vitamine B3, soit en association avec elle, \u00bb explique le professeur Linster. \u00ab En continuant \u00e0 explorer les m\u00e9canismes mol\u00e9culaires de la maladie dans des cultures cellulaires en laboratoire, nous am\u00e9liorons non seulement notre compr\u00e9hension des processus enzymatiques impliqu\u00e9s mais nous ouvrons \u00e9galement la voie vers des traitements plus efficaces.&nbsp;\u00bb<\/p>\n\n\n\n<p>Les petites mol\u00e9cules identifi\u00e9es gr\u00e2ce \u00e0 ces recherches men\u00e9es en collaboration avec des scientifiques australiens et des coll\u00e8gues du LCSB, notamment l\u2019\u00e9quipe&nbsp;<a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/developmental-cellular-biology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Developmental and Cellular Biology<\/a>&nbsp;et la&nbsp;<a href=\"https:\/\/www.uni.lu\/lcsb-en\/facilities\/scientific-platforms\/metabolomics\/\" target=\"_blank\" rel=\"noreferrer noopener\">plateforme m\u00e9tabolomique &amp; lipidomique<\/a>, constituent en effet des candidats prometteurs pour diff\u00e9rentes maladies neuro-m\u00e9taboliques rares. Celles-ci peuvent entra\u00eener une carence en vitamine B12, elle-m\u00eame associ\u00e9e \u00e0 certaines maladies neurod\u00e9g\u00e9n\u00e9ratives, en particulier chez les personnes ayant un faible apport alimentaire en vitamine.<br><br>Le professeur Carole Linster et son \u00e9quipe ont d\u00e9couvert que ce g\u00e8ne code pour une enzyme, appel\u00e9e CLYBL, impliqu\u00e9e elle aussi dans l\u2019\u00e9limination de m\u00e9tabolites toxiques. CLYBL d\u00e9grade un compos\u00e9 baptis\u00e9 Malyl-CoA, assurant ainsi le bon fonctionnement de processus cellulaires li\u00e9 \u00e0 la vitamine B12.<\/p>\n\n\n\n<ul class=\"wp-block-unilux-blocks-custom-buttons btn-list\"><\/ul>\n<\/div>\n<\/div>\n<\/div>\n<\/section>\n\n\n<section class=\"py-0 wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>Les scientifiques ont ensuite explor\u00e9 en d\u00e9tail les m\u00e9canismes reliant CLYBL, Malyl-CoA et carence en vitamine B12. Au cours de cette nouvelle \u00e9tude <a href=\"https:\/\/www.nature.com\/articles\/s41589-025-01857-9\" target=\"_blank\" rel=\"noreferrer noopener\">publi\u00e9e dans la revue <em>Nature Chemical Biology<\/em><\/a>, ils ont pour la premi\u00e8re fois d\u00e9tect\u00e9 Malyl-CoA dans des cellules humaines et ont montr\u00e9 qu&rsquo;il s&rsquo;agit d&rsquo;un sous-produit du cycle de Krebs, un processus vital pour convertir les aliments en \u00e9nergie. Au-del\u00e0 de l\u2019avanc\u00e9e scientifique, leurs r\u00e9sultats mettent en lumi\u00e8re que le savoir-faire acquis en \u00e9tudiant les maladies rares peut apporter un \u00e9clairage pour des pathologies plus courantes.<\/p>\n\n\n\n<p>En effet, une mutation sur le g\u00e8ne CLYBL qui entra\u00eene un dysfonctionnement de l&rsquo;enzyme appara\u00eet dans un pourcentage anormalement \u00e9lev\u00e9 de la population. \u00ab Bien qu&rsquo;apparemment pr\u00e9judiciable en raison de son lien avec la carence en vitamine B12, cette mutation pourrait avoir procur\u00e9 un avantage \u00e9volutif, expliquant pourquoi elle est aujourd\u2019hui plus fr\u00e9quente dans la population. Elle pourrait par exemple contribuer \u00e0 limiter la virulence des bact\u00e9ries pathog\u00e8nes qui d\u00e9pendent de cette vitamine pour leur croissance et leur survie, \u00bb souligne le professeur Linster. \u00ab Nos r\u00e9sultats remettent en question l&rsquo;id\u00e9e selon laquelle les d\u00e9faillances au niveau des m\u00e9canismes de r\u00e9paration et d\u2019\u00e9limination des m\u00e9tabolites ne concernent que des maladies rares. font-family: &quot;object-fit: cover; object-position: 50.00% 65.00%;&quot;; aspect-ratio: 16\/9; object-fit: cover; width: 100%;\"\n        loading=\"lazy\"\n\/>    <\/figure><\/li>        <\/ul>\n\n        <!-- Swiper pagination -->\n        <div class=\"swiper-pagination\">\n            <div class=\"swiper-pagination__bullets\"><\/div>\n        <\/div>\n    <\/div>\n<\/section><\/div>\n\n\n\n<div class=\"wp-block-unilux-blocks-wrapper w-full lg:w-5\/12 lg:order-1 lg:px-32\">\n<h3 class=\"has-text-align-left wp-block-unilux-blocks-heading\"    >\n<\/h3>\n\n\n\n<p>Au-del\u00e0 de ses travaux de recherche, le LCSB souhaite sensibiliser le grand public au sujet des maladies rares.<br><br>Lors de l\u2019\u00e9dition 2024 des&nbsp;<a href=\"https:\/\/researchersdays.science.lu\/fr\" target=\"_blank\" rel=\"noreferrer noopener\">Researchers\u2019 Days<\/a>, une f\u00eate de la science organis\u00e9e tous les deux ans par le&nbsp;<a href=\"https:\/\/www.fnr.lu\/\" target=\"_blank\" rel=\"noreferrer noopener\">Luxembourg National Research Fund<\/a>&nbsp;(FNR), le groupe Enzymology and Metabolism a par exemple organis\u00e9 un atelier interactif sur le sujet. Des centaines de visiteurs de tous \u00e2ges ont ainsi pu d\u00e9crypter le code g\u00e9n\u00e9tique, faire des exp\u00e9riences avec des enzymes et d\u00e9couvrir comment les maladies rares sont \u00e9tudi\u00e9es gr\u00e2ce \u00e0 des cultures cellulaires et des poissons z\u00e8bres ou diagnostiqu\u00e9es chez les enfants.<\/p>\n\n\n\n<ul class=\"wp-block-unilux-blocks-custom-buttons btn-list\"><\/ul>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<\/section>\n\n\n<section class=\"py-0 wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>Poursuivant la tradition entam\u00e9e l&rsquo;ann\u00e9e derni\u00e8re, lorsque le b\u00e2timent Biotech II situ\u00e9 sur le campus de Belval a \u00e9t\u00e9 illumin\u00e9 pour la premi\u00e8re fois aux couleurs de la journ\u00e9e mondiale, le LCSB est engag\u00e9 dans la lutte contre les maladies rares sur tous les fronts&nbsp;: en proposant des activit\u00e9s de sensibilisation, en d\u00e9veloppant de collaborations nationales et internationales avec l\u2019ensemble de la communaut\u00e9 des maladies rares et en assurant une recherche de pointe sur les m\u00e9canismes pathologiques et les approches th\u00e9rapeutiques.<\/p>\n\n\n\n<p>\u00c0 cette fin, le LCSB a r\u00e9cemment cr\u00e9\u00e9 un&nbsp;<a href=\"https:\/\/www.uni.lu\/lcsb-fr\/soutenez-nous\/fonds-maladies-rares-enfant\/\" target=\"_blank\" rel=\"noreferrer noopener\">fonds<\/a>&nbsp;afin de soutenir la recherche sur les maladies infantiles rares. Tous les dons collect\u00e9s via ce fonds contribueront \u00e0 acc\u00e9l\u00e9rer les avanc\u00e9es scientifiques, apportant de l&rsquo;espoir aux familles concern\u00e9es.
