En outre, elles pr\u00e9sentent souvent des sympt\u00f4mes plus graves. Toutefois, les m\u00e9canismes \u00e0 l&rsquo;origine de ces diff\u00e9rences ne sont pas encore bien compris. Une nouvelle \u00e9tude vient cependant de mettre en \u00e9vidence des changements \u00e0 l\u2019\u00e9chelle mol\u00e9culaire dans plusieurs types de cellules du cerveau qui diff\u00e8rent entre hommes et femmes atteints de la maladie d&rsquo;Alzheimer, sugg\u00e9rant de nouvelles cibles pour des approches th\u00e9rapeutiques personnalis\u00e9es. L&rsquo;\u00e9tude, dirig\u00e9e par le <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/enrico-glaab\/\">professeur Enrico Glaab<\/a> du <a href=\"https:\/\/www.uni.lu\/lcsb-en\/\">Luxembourg Centre for Systems Biomedicine<\/a> (LCSB) de l&rsquo;Universit\u00e9 du Luxembourg, a \u00e9t\u00e9 publi\u00e9e r\u00e9cemment dans <a href=\"https:\/\/alz-journals.onlinelibrary.wiley.com\/doi\/10.1002\/alz.14476\"><em>Alzheimer&rsquo;s &amp; Dementia<\/em><\/a>.<\/p>\n\n\n\n<h2 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"zoomer-cellule-par-cellule\"\n    >\n<strong><strong>Zoomer cellule par cellule<\/strong><\/strong><\/h2>\n\n\n\n<p>L&rsquo;\u00e9quipe a r\u00e9alis\u00e9 des analyses pouss\u00e9es, bas\u00e9es sur la technologie \u00ab single cell \u00bb, pour plus de 2,3 millions de cellules c\u00e9r\u00e9brales, dont des neurones et diff\u00e9rents types de cellules gliales, provenant de personnes atteintes de la maladie d&rsquo;Alzheimer et de t\u00e9moins sains ayant fait don de leur cerveau \u00e0 la recherche apr\u00e8s leur mort. Les chercheurs se sont plus particuli\u00e8rement int\u00e9ress\u00e9s au cortex pr\u00e9frontal, une zone cl\u00e9 responsable de la prise de d\u00e9cision et de la r\u00e9cup\u00e9ration des souvenirs. Ils ont mesur\u00e9 les niveaux d&rsquo;expression de milliers de g\u00e8nes dans chaque cellule, un processus \u00e9galement appel\u00e9 transcriptomique en cellule unique. Ils ont ensuite compar\u00e9 patients et t\u00e9moins sains mais aussi hommes et femmes afin de rechercher des changements li\u00e9s \u00e0 la maladie dans chacun des sexes. Cette approche innovante leur a permis d&rsquo;analyser avec une pr\u00e9cision sans pr\u00e9c\u00e9dent l&rsquo;activit\u00e9 des g\u00e8nes et les processus cellulaires dans plusieurs types de cellules c\u00e9r\u00e9brales impliqu\u00e9s dans la maladie d&rsquo;Alzheimer.<\/p>\n\n\n\n<p>Leurs r\u00e9sultats ont r\u00e9v\u00e9l\u00e9 des signatures mol\u00e9culaires distinctes chez les hommes et les femmes atteints de la maladie d&rsquo;Alzheimer. Chez les patients de sexe masculin, les chercheurs ont observ\u00e9 des changements significatifs dans les voies associ\u00e9es \u00e0 la mort cellulaire au niveau des astrocytes, un sous-type de cellules gliales responsables des fonctions m\u00e9taboliques, structurelles, hom\u00e9ostatiques et neuroprotectrices. Les patientes pr\u00e9sentaient en revanche des alt\u00e9rations des voies impliqu\u00e9es dans la communication cellulaire et la r\u00e9gulation de la croissance. \u00ab En nous pla\u00e7ant \u00e0 l\u2019\u00e9chelle de la cellule unique, nous pouvons \u00e9tudier les m\u00e9canismes pathologiques d\u00e9pendants du sexe et les diff\u00e9rences subtiles entre les divers types de cellules du cerveau qui pourraient autrement rester cach\u00e9es, \u00bb explique <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/mohamed-soudy\/\">Mohamed Soudy<\/a>, doctorant au sein du <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/biomedical-data-science\/\">groupe Biomedical Data Science<\/a> du LCSB et premier auteur de l&rsquo;\u00e9tude.<\/p>\n\n\n\n<h2 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"differences-entre-hommes-et-femmes-au-niveau-de-la-communication-cellulaire\"\n    >\n<strong><strong>Diff\u00e9rences entre hommes et femmes au niveau de la communication cellulaire<\/strong><\/strong><\/h2>\n\n\n\n<p>L&rsquo;une des principales caract\u00e9ristiques de la maladie d&rsquo;Alzheimer est la d\u00e9t\u00e9rioration de la communication entre les cellules c\u00e9r\u00e9brales qui entra\u00eene la mort des neurones et une d\u00e9gradation g\u00e9n\u00e9ralis\u00e9e des fonctions c\u00e9r\u00e9brales. Les observations des chercheurs indiquent que ce processus se manifeste diff\u00e9remment selon le sexe. Les cerveaux des patients pr\u00e9sentaient des changements prononc\u00e9s dans la signalisation de la mort cellulaire, tandis que les cerveaux des patientes montraient une alt\u00e9ration de la signalisation du calcium, un processus cl\u00e9 dans les fonctions c\u00e9r\u00e9brales et la m\u00e9moire. \u00ab Ces modifications propres \u00e0 chacun des sexe et les interactions complexes entre les diff\u00e9rentes processus cellulaires permettent de mieux comprendre pourquoi la maladie d&rsquo;Alzheimer se pr\u00e9sente diff\u00e9remment chez les hommes et les femmes, \u00bb explique <a href=\"https:\/\/www.uni.lu\/lcsb-en\/people\/sophie-le-bars\/\">Dr Sophie LeBars<\/a>, post-doctorante et coauteur de l&rsquo;\u00e9tude.<\/p>\n\n\n\n<h2 class=\"has-text-align-left wp-block-unilux-blocks-heading\"        id=\"cibler-les-mecanismes-propres-a-chaque-sexe-via-des-traitements-adaptes\"\n    >\n<strong><strong>Cibler les m\u00e9canismes propres \u00e0 chaque sexe via des traitements adapt\u00e9s<\/strong><\/strong><\/h2>\n\n\n\n<p>Pour rechercher de potentielles cibles sur lesquelles des m\u00e9dicaments pourraient agir, les chercheurs ont utilis\u00e9 des mod\u00e8les informatiques qui d\u00e9crivent comment les r\u00e9gulateurs mol\u00e9culaires interagissent entre eux et avec d&rsquo;autres substances dans la cellule pour r\u00e9gir les niveaux d&rsquo;expression des g\u00e8nes. L&rsquo;analyse syst\u00e9matique de ces r\u00e9seaux de r\u00e9gulation g\u00e9nique pour diff\u00e9rents types de cellule et la simulation des effets potentiels des m\u00e9dicaments ont permis de rep\u00e9rer des mol\u00e9cules cl\u00e9s. Les cibler pourraient permettre d&rsquo;inverser les changements pathologiques propres \u00e0 chaque sexe. Chez les patients de sexe masculin, RPTOR, une mol\u00e9cule impliqu\u00e9e dans la survie et le m\u00e9tabolisme des cellules, a \u00e9t\u00e9 identifi\u00e9e comme un m\u00e9diateur potentiel des changements li\u00e9s \u00e0 la maladie d\u2019Alzheimer. Chez les femmes, les chercheurs ont identifi\u00e9 LRP1 comme une mol\u00e9cule cl\u00e9. Elle joue notamment un r\u00f4le important dans l&rsquo;\u00e9limination des prot\u00e9ines toxiques, telles que l&rsquo;amylo\u00efde-b\u00eata, du cerveau.
