{"id":2542,"date":"2023-10-24T15:06:33","date_gmt":"2023-10-24T13:06:33","guid":{"rendered":"https:\/\/www.uni.lu\/lcsb-en\/?post_type=news&#038;p=2542"},"modified":"2024-08-09T11:26:09","modified_gmt":"2024-08-09T09:26:09","slug":"gut-brain-axis-when-diet-and-microbiome-modulate-parkinson","status":"publish","type":"news","link":"https:\/\/www.uni.lu\/lcsb-en\/news\/gut-brain-axis-when-diet-and-microbiome-modulate-parkinson\/","title":{"rendered":"Gut-brain axis: When diet and microbiome modulate Parkinson\u2019s disease progression"},"content":{"rendered":"\n<section class=\"wp-block-unilux-blocks-free-section section\"><div class=\"container xl:max-w-screen-xl\">\n<p>In a recent study, two teams from the <a href=\"https:\/\/www.uni.lu\/lcsb-en\/\" target=\"_blank\" rel=\"noreferrer noopener\">Luxembourg Centre for Systems Biomedicine<\/a> (LCSB) of the University of Luxembourg, as well as national and international partners, collaborated to explore the role of diet and the gut microbiome in the progression of Parkinson\u2019s disease. Their results, <a href=\"https:\/\/www.cell.com\/cell-reports\/fulltext\/S2211-1247(23)01082-3\" target=\"_blank\" rel=\"noreferrer noopener\">published in the scientific journal Cell Reports<\/a>, show that dietary fibre deprivation and exposure to a specific bacterial protein can exacerbate the disease, highlighting the importance of lifestyle management for people with Parkinson\u2019s disease.<\/p>\n\n\n\n<p>Parkinson\u2019s disease is a neurological disorder characterised by the loss of specific neurons in the brain and the aggregation of a human protein called \u03b1-synuclein. While several risk factors for this disease are well-known, it is not yet clear what can modulate its progression over time. In this context, researchers from the <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/systems-ecology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Systems Ecology<\/a> group and the <a href=\"https:\/\/www.uni.lu\/lcsb-en\/research-groups\/neuropathology\/\" target=\"_blank\" rel=\"noreferrer noopener\">Neuropathology<\/a> group at the LCSB combined their respective expertise to study the effects of diet, the gut microbiome\u2019s composition and proteins secreted by these microorganisms in a mouse model of Parkinson\u2019s disease.<\/p>\n\n\n\n<p>\u201cBased on a series of tests, we determined that a specific transgenic mouse line, that replicates key features of Parkinson\u2019s disease, was an appropriate model to investigate the impact of environmental factors, such as diet, on the evolution of this disease,\u201d explains <a href=\"https:\/\/www.medizin.uni-tuebingen.de\/de\/das-klinikum\/mitarbeiter\/profil\/5116\" target=\"_blank\" rel=\"noreferrer noopener\">Dr Kristopher Schmit<\/a>, who conducted the study during his PhD within the Systems Ecology and the Neuropathology groups. \u201cThese mice overexpress the protein \u03b1-synuclein which leads to the progressive apparition of motor symptoms, just like in patients.\u201d<\/p>\n\n\n\n<p>When these transgenic mice were fed a diet with low amounts of fibre, the microbial diversity in their gut decreased. In particular, bacteria associated with neuroprotective functions declined, whereas mucus-foraging ones increased, leading to a thinning of the outer mucus layer in the colon. \u201cAs the mucus layers act as a physical barrier against pathogens, mucus erosion driven by changes in the microbiome could facilitate access of pathogenic agents to the deeper layers of the gut, notably reaching its nerve cells,\u201d details <a href=\"https:\/\/www.uni.lu\/en\/person\/NTAwMDMzMzVfX1BhdWwgV0lMTUVT\/\" target=\"_blank\" rel=\"noreferrer noopener\">Prof. Paul Wilmes<\/a>, head of the Systems Ecology group.<\/p>\n\n\n\n<p>Additionally, when combining fibre deprivation with the exposure to curli, a protein produced by bacteria such as Escherichia coli and which can accelerate the aggregation of human \u03b1-synuclein, the gut barrier integrity was further disrupted. Similarly, \u03b1-synuclein aggregation in the brain intensified with the presence of curli in the gut and increased further with fibre deprivation. \u201cMoreover, when assessing the loss of neurons in the brain of these mice, we concluded that curli combined with a low amount of fibre drove neurodegeneration in this model of Parkinson\u2019s disease,\u201d explains <a href=\"https:\/\/www.uni.lu\/en\/person\/NTAwMDExNDZfX01hbnVlbCBCVVRUSU5J\/\" target=\"_blank\" rel=\"noreferrer noopener\">Dr Manuel Buttini<\/a>, senior researcher in the Neuropathology group led by Prof. Michel Mittelbronn.<\/p>\n\n\n\n<p>Taken together, these results show that, to different extents, diet and exposure to bacterial proteins produced by the microbiome worsen motor performance as well as intestinal and brain pathologies. The authors outline the possible mechanism at play: \u201cTo explain our findings, we propose the following sequence of events, whereby chronic dietary fibre deprivation leads to changes in microbial populations, also called dysbiosis. This then sparks gut mucus erosion and increased gut permeability, facilitating entry of pathogenic agents. In particular, the resulting higher exposure of enteric neurons to proteins such as curli causes greater \u03b1-synuclein aggregation in the enteric and central nervous system, leading to neurodegeneration and exacerbation of motor deficits.\u201d<\/p>\n\n\n\n<p>The study sheds important insights on how an environmental factor such as diet and its action through the gut microbiome can modulate Parkinson\u2019s disease progression via the gut-brain axis. It also has implications for lifestyle adjustments that could mitigate the evolution of the disease, such as ensuring an optimal balanced diet and oversight of the use of antibiotics that could favour the proliferation of harmful bacteria like those expressing curli.<\/p>\n\n\n\n<p>&#8212;<\/p>\n\n\n\n<p><strong>Reference:<\/strong> Kristopher J.
